Armodafinil
The long-duration eugeroic. A complete guide to the purified (R)-enantiomer of modafinil — mechanism, dosing protocols, and why it is not the same compound as racemic modafinil.
What is Armodafinil?
Armodafinil is the purified (R)-enantiomer of modafinil — one half of the racemic pair, isolated on its own. It is a eugeroic (wakefulness-promoting) agent sold under the brand name Nuvigil and FDA-approved in 2007 for excessive daytime sleepiness associated with narcolepsy, obstructive sleep apnea, and shift work disorder. Because the fast-clearing (S)-enantiomer has been removed, armodafinil holds higher, steadier plasma levels into the afternoon and evening than the same milligrams of racemic modafinil. The two compounds are close relatives — but they are not the same drug, and they are not interchangeable mg-for-mg.

Armodafinil vs Modafinil
Same family, different compound. Racemic modafinil is a 50/50 blend of two mirror-image molecules; armodafinil is only the long-lasting one. That single difference changes the dose, the curve, and the duration.
| Feature | Modafinil (Provigil) | Armodafinil (Nuvigil) |
|---|---|---|
| Composition | Racemic mixture — 50% (R) + 50% (S) enantiomers | Pure (R)-enantiomer only |
| Half-Life | (S)-enantiomer clears in ~4 hours; (R) in ~15 hours | ~15 hours |
| Plasma Curve | Levels fall through the day as the (S) half clears | Higher, steadier levels later in the day at equal mg |
| Typical Dose | 200mg once each morning | 150mg once each morning (250mg in narcolepsy / sleep apnea) |
| FDA Approval | 1998 — narcolepsy, sleep apnea, shift work disorder | 2007 — the same three indications |
| Legal Status | Schedule IV controlled substance | Schedule IV controlled substance |
| The Bottom Line | Not interchangeable mg-for-mg. 150mg of armodafinil is not "150mg of modafinil" — it lasts longer and holds higher late-day levels, so protocols are dosed and timed differently. | |
Administration Protocols
Armodafinil is a synthetic small-molecule compound supplied as oral tablets. It does not require bacteriostatic water or injection. Because its ~15-hour half-life reaches deep into the evening, timing is the most critical factor in the protocol.
Form Factor 💊
Armodafinil is researched in 50mg, 150mg, 200mg, and 250mg oral tablets. The 150mg strength is the typical approved dose; the range exists so protocols can start low and step up deliberately.
Strict Timing ⏱️
Armodafinil has a ~15 hour half-life — the longest of the afinil family. It is taken once daily, immediately upon waking. For shift-work protocols, take it about 1 hour before the shift begins. A dose taken late in the morning will still be active at bedtime.
Synergy & Hydration 💧
Like modafinil, armodafinil increases choline utilization in the brain, and researchers often pair it with a choline source (like Alpha-GPC) to head off headaches. It also suppresses the sensation of thirst, so deliberate water intake is mandatory.
Research Dosages
The Half-Life Rule
Do not underestimate the duration of this compound. A 150mg dose taken at 12:00 PM (Noon) still has roughly half its strength circulating at Midnight, making natural sleep nearly impossible. Morning dosing is not a suggestion — it is the protocol.
| Protocol Goal | Dosage | Frequency / Timing | Notes |
|---|---|---|---|
| Cognitive Enhancement (Beginner / Sensitive) |
50mg | Early AM | The lowest tablet strength. A gentle introduction that favors clean alertness over intensity — the right first protocol. |
| Standard Wakefulness (Typical Approved Dose) |
150mg | Early AM | The most commonly prescribed strength for narcolepsy, obstructive sleep apnea, and shift work disorder. |
| Maximum Wakefulness (Narcolepsy / Sleep Apnea) |
250mg | Early AM | The top approved dose, used in narcolepsy and sleep apnea protocols. Expect the strongest effect and the longest tail. |
| Shift Work Protocol | 150mg | Pre-Shift | Taken about 1 hour before a night shift starts. Plan the ~15-hour tail so it winds down before daytime sleep. |
| Cycle Frequency | N/A | 2-3x Per Week | Approved medical regimens dose daily, but cognitive research protocols commonly cycle 2–3x per week to limit tolerance and protect sleep architecture. |
Safety, Interactions & Cautions
Common effects: headache, nausea, insomnia, dizziness, anxiety, and dry mouth — generally mild and dose-related, and the reason the lowest effective dose is the right dose.
Stop at the first sign of a rash. Rare but serious skin reactions, including Stevens–Johnson syndrome, have been reported with this compound class. Any rash means discontinue immediately and seek medical care — do not dose through it.
Hormonal contraception: armodafinil induces the CYP3A4 enzyme and lowers blood levels of hormonal birth control. Use an alternative or additional contraceptive method during use and for one month after the last dose.
Other medications: armodafinil inhibits CYP2C19, which raises exposure to drugs cleared by that pathway — omeprazole, diazepam, and phenytoin among them. Doses of those medications may need adjusting.
Mind, mood & heart: use caution with any history of mania or psychosis, which wakefulness agents can aggravate, and with uncontrolled high blood pressure.
Controlled status: armodafinil is a Schedule IV controlled substance — lower abuse liability than classic amphetamine stimulants, but a controlled substance all the same, and handled accordingly.
Storage & Handling
Temperature
Armodafinil tablets are highly stable at room temperature (68°F to 77°F). Do not refrigerate.
Moisture
Keep tablets in their original blister packs until the exact moment of use to prevent moisture degradation.
Light Exposure
Store in a cool, dark place (like a drawer or medicine cabinet) away from direct UV sunlight.
