Peptopia Research Report
GLP-1s & Biological Aging
The Longevity Report
โง โง โง
Fox Business is covering a striking new chapter in the GLP-1 story. At the Aging Research & Drug Discovery conference, Eli Lilly and Novo Nordisk — the makers of Mounjaro/Zepbound and Ozempic/Wegovy — presented research suggesting their drugs may slow, or even partially reverse, biological aging. This report lays out what was actually presented, how scientists measure “age,” the benefits being reported, and the honest caveats.

๐ฏ The headline: two to three years younger?
The claim now making the rounds on business television: the world’s best-selling weight-loss drugs may also be longevity drugs. At the Aging Research & Drug Discovery (ARDD) conference, both Eli Lilly and Novo Nordisk presented analyses from their clinical-trial programs indicating that GLP-1 medications slowed biological aging by roughly two to three years in the patients studied.
Geneticist Steve Horvath — inventor of the “Horvath clock,” the original epigenetic age test — called two years “a pretty strong effect” and said the data suggests GLP-1 drugs may be geroprotectors: medications that slow, or possibly even reverse, biological aging.
For scale: more than one in ten American adults now takes a GLP-1 for weight loss. If even part of the aging signal holds up, the implications go far beyond the scale.
๐ฌ The numbers — Novo Nordisk & Eli Lilly
10,052
patients in Novo Nordisk’s analysis — half on semaglutide, half on placebo, blood drawn at baseline and months later
Proteomic clocks · ARDD 2026
โ2 to โ4 yrs
reduction in heart age for patients on semaglutide vs. control — the strongest signal in either dataset
Novo Nordisk · proteomic clock
โ1 to โ4 yrs
reduction in age of kidney, brain, lung, and pancreas on semaglutide
Novo Nordisk · proteomic clocks
9%
slower pace of biological aging on semaglutide in the UC San Diego study (DunedinPACE clock), published in Nature Communications
UC San Diego · adults with HIV · 2026
Eli Lilly’s data told a similar story on a smaller scale. Using epigenetic clocks — which read chemical marks on DNA rather than blood proteins — Lilly’s tirzepatide analysis showed the clearest age reductions in the heart, brain, liver, and kidneys.
“All the clocks are telling a consistent story that we’re seeing a reduction in age.”— Kevin Duffin, VP of aging research, Eli Lilly, at ARDD 2026
Duffin added a note of proportion: this is a meaningful reduction, not decades — nobody is claiming thirty years. The effects varied widely depending on which clock was used and which organ was measured — up to four years on a heart-protein clock, less elsewhere — but every clock pointed the same direction.
๐งฌ How scientists measure biological age
You can’t ask a birth certificate how fast someone is aging. Researchers use biological-age clocks — algorithms that estimate how old your cells and organs act, versus how old you are.
๐ฐ๏ธ Epigenetic clocks (Horvath clocks)
Read DNA methylation marks — chemical tags on your genes that accumulate with age. Lilly used these for its tirzepatide analysis. The original Horvath clock (2013) kicked off the entire field.
๐ฉธ Proteomic clocks
Read levels of key proteins in the blood to predict a person’s age and risk of dying. Novo Nordisk used these across its 10,000-patient analysis.
โฑ๏ธ DunedinPACE — the pace-of-aging clock
Instead of a single age number, measures the speed of aging. The UC San Diego team found semaglutide slowed that pace by 9% — in adults with HIV, a population that ages faster than average.
Why it matters: when independent clock types — DNA marks, blood proteins, pace-of-aging — all move the same direction, researchers take notice. That convergence is what made the ARDD presentations land.
โจ The benefits being reported
The segment walked through a long list. Here it is, organized by how established each benefit is:
โ๏ธ Weight loss
The original breakthrough — the reason these drugs exist.
Established
๐ฉบ Blood sugar control
Core diabetes indication for both semaglutide and tirzepatide.
Established
โค๏ธ Heart age (โ2 to โ4 yrs)
Strongest signal in Novo’s proteomic analysis; echoed in Lilly’s data.
New data
๐ง ๐งช Organ age (โ1 to โ4 yrs)
Kidney, brain, lung, pancreas (Novo); heart, brain, liver, kidney (Lilly).
New data
๐ Blood pressure
Improvements seen through weight loss and weight-independent mechanisms.
Established
๐งช Cholesterol
Lipid improvements reported across multiple GLP-1 trials.
Established
๐ฅ Inflammation
Markers of systemic inflammation trend down — one suspected aging pathway.
Emerging
๐ถ Physical function
Early signals on mobility and walking speed; being tested directly in new trials.
Emerging
๐ซ Whole-body outcomes map
Oct 28: 70,982 matched patients, 181 outcomes — lower heart attack (HR 0.53), heart failure, and kidney risk on GLP-1s vs older diabetes drugs (JDST).
New data
โป๏ธ Defending surgical weight loss
Oct: after bariatric surgery, GLP-1 users lost 14.8% of regained weight at 12 months vs 8.1% for nonusers, no added complication signal (Obesity).
New data
โ ๏ธ The honest caveats
- Company-presented data. Both datasets were presented by the drugmakers themselves at a conference — not yet peer-reviewed publications. Independent replication is pending.
- The population studied. The analyses covered overweight or diabetic patients. Whether healthy people see the same anti-aging effects is unknown — that’s exactly what the new ARPA-H trial aims to find out.
- Muscle loss is a real concern. In older adults, appetite suppression can cut protein intake and accelerate muscle loss — researchers flag this as the key risk to manage.
- Side effects exist. GI effects are common; rare but serious risks have been reported. These drugs are not approved for anti-aging, and nothing here is a recommendation to take them for that purpose.
- Clocks aren’t destiny. A younger proteomic age is a biomarker signal, not proof of extra years of life. Longevity has to be proven the slow way — with time.
- Joints & bones (Oct 28). The 181-outcome study also found higher osteoarthritis (HR 1.48) and stress-fracture (HR 1.68) associations on GLP-1s — observational signals, not proof of cause.
๐ญ What’s next
๐งช UTMB Galveston — the first dedicated trial
University of Texas Medical Branch researchers are running one of the first US clinical trials testing a GLP-1 specifically for aging: adults 55–70 on tirzepatide for 24 weeks, then 12 weeks of follow-up, tracking epigenetic clocks plus physical function.
๐๏ธ ARPA-H — $38 million bet
The US Advanced Research Projects Agency for Health is funding a study of semaglutide in healthy adults over 60, measuring cognition, mobility, and sensory acuity — the outcomes that actually matter for aging well.
๐ฏ Retatrutide — the triple agonist
Nov 2, 2026: first phase 3 head-to-head win — beat Ozempic 2 mg on A1C (−2.2 vs −1.8) and weight (−20.9% vs −9.4%), TRANSCEND-T2D-2 topline. Investigational; next test is TRIUMPH-5 vs tirzepatide. Read the story.
๐ The oral wave: orforglipron beat oral semaglutide head-to-head, showed modeled diabetes/CV risk cuts, and matched insulin on heart safety — as Biomea’s BMF-650 posted Phase 1 signals.
Fox News senior medical analyst Dr. Marc Siegel has also covered the UC San Diego findings on air, noting the cardiovascular and neurological angles alongside the digestive and thyroid side-effect profile — a sign this story is moving from conference halls to mainstream medicine’s radar.
