Retatrutide Guide
The highly advanced "Triple Agonist". A complete guide to mechanism, reconstitution, and standard phase 3 titration protocols.
What is Retatrutide?
Retatrutide is an investigational, next-generation peptide known as a triple hormone receptor agonist. By combining GLP-1, GIP, and Glucagon receptor agonism into a single molecule, it produces unprecedented synergistic effects on metabolic regulation, energy expenditure, and fat reduction.
GLP-1 Agonist
๐ Signals strong satiety
โณ Delays gastric emptying
๐ Suppresses appetite
GIP Agonist
๐ Enhances insulin sensitivity
๐ก๏ธ Buffers GLP-1 nausea
๐ฉธ Regulates blood glucose
Glucagon (GCG)
๐ฅ Spikes energy expenditure
โก Direct fat burning (lipolysis)
๐ซ Clears liver fat
Reconstitution Basics
Lyophilized (freeze-dried) Retatrutide must be reconstituted with Bacteriostatic Water (BAC) before research use. Follow these standard sterile handling protocols:
Prep & Sanitize ๐งผ
Wash hands thoroughly. Pop the plastic caps off both the Retatrutide vial and the BAC water vial. Swab both rubber stoppers with an alcohol prep pad and let air dry.
Draw BAC Water ๐
Using a sterile mixing syringe, draw the required amount of Bacteriostatic Water. (e.g., Adding 1mL of BAC to a 10mg vial yields a concentration of 1mg per 0.1mL/10 units).
Inject & Dissolve ๐ง
Pierce the peptide vial. Do not spray directly onto the powder. Angle the needle so the water drips slowly down the glass wall of the vial to protect the fragile peptide bonds.
Swirl Gently ๐
NEVER SHAKE. Gently roll or swirl the vial between your fingers until the solution is completely clear and dissolved. It is now ready for research.
Standard Titration Schedule
Based on Phase 3 clinical trials, Retatrutide requires a meticulous step-up escalation schedule administered once every 7 days. Slow escalation is mandatory to allow the body to adjust to the powerful triple-receptor activation and avoid severe nausea ๐คข.
| Timeline | Weekly Dose | Phase | Goal / Notes |
|---|---|---|---|
| Weeks 1 - 4 | 2.0 mg | Initiation Phase | Acclimate the GI tract; strictly for building initial tolerance. |
| Weeks 5 - 8 | 4.0 mg | Escalation Phase | Initial dose escalation. Mild therapeutic effects and fat oxidation begin. |
| Weeks 9 - 12 | 6.0 mg | Escalation Phase | Smoothed escalation step to prevent severe GI shock before higher doses. |
| Weeks 13 - 16 | 9.0 mg | Therapeutic Phase | Standard high-therapeutic dose. Significant metabolic changes expected. |
| Weeks 17+ | 12.0 mg | Maintenance / Max | Maximum clinical dose. Maintained long-term for robust research outcomes. |
Storage & Handling
Temperature
Store in the refrigerator (36ยฐF to 46ยฐF) both before and after reconstitution. Do not freeze.
Light Exposure
Peptides are highly sensitive to UV light. Keep vials in their boxes or a dark container.
Shelf Life
Once mixed with BAC water, discard the vial after 28 days to ensure sterility and efficacy.
