Semaglutide Guide
The foundational GLP-1 receptor agonist. A complete guide to mechanism, reconstitution, and standard titration protocols.
What is Semaglutide?
Semaglutide is a highly effective, long-acting synthetic peptide that mimics the human glucagon-like peptide-1 (GLP-1) hormone. By binding selectively to GLP-1 receptors, it initiates a powerful trifecta of physiological changes that regulate metabolism and appetite.
Neurological
๐ง Crosses blood-brain barrier
๐ Signals strong satiety
๐ Reduces food noise
Gastrointestinal
โณ Delays gastric emptying
๐ฝ๏ธ Prolongs physical fullness
๐ก๏ธ Reduces overall intake

Reconstitution Basics
Lyophilized (freeze-dried) Semaglutide must be reconstituted with Bacteriostatic Water (BAC) before research use. Follow these standard sterile handling protocols:
Prep & Sanitize ๐งผ
Wash hands thoroughly. Pop the plastic caps off both the Semaglutide vial and the BAC water vial. Swab both rubber stoppers with an alcohol prep pad and let air dry.
Draw BAC Water ๐
Using a sterile mixing syringe, draw the required amount of Bacteriostatic Water. (e.g., Adding 1mL of BAC to a 2mg vial yields a concentration of 0.2mg per 0.1mL/10 units).
Inject & Dissolve ๐ง
Pierce the peptide vial. Do not spray directly onto the powder. Angle the needle so the water drips slowly down the glass wall of the vial to protect the fragile peptide bonds.
Swirl Gently ๐
NEVER SHAKE. Gently roll or swirl the vial between your fingers until the solution is completely clear and dissolved. It is now ready for research.
Standard Titration Schedule
Because Semaglutide profoundly impacts gastric emptying, researchers must follow a 5-step titration schedule, administering the peptide once every 7 days to allow the body to adjust and avoid severe nausea ๐คข.
| Timeline | Weekly Dose | Phase | Goal / Notes |
|---|---|---|---|
| Weeks 1 - 4 | 0.25 mg | Initiation Phase | Acclimate the body; strictly for building initial tolerance. |
| Weeks 5 - 8 | 0.50 mg | Titration Phase | Initial dose escalation. Mild therapeutic effects may begin. |
| Weeks 9 - 12 | 1.00 mg | Therapeutic Phase | Standard therapeutic dose. Significant appetite suppression expected. |
| Weeks 13 - 16 | 1.70 mg | Therapeutic Phase | Advanced therapeutic tier. Only advance if 1.0mg efficacy has stalled. |
| Weeks 17+ | 2.40 mg | Maintenance / Max | Maximum clinical dose. Maintained long-term for robust research outcomes. |
Storage & Handling
Temperature
Store in the refrigerator (36ยฐF to 46ยฐF) both before and after reconstitution. Do not freeze.
Light Exposure
Peptides are highly sensitive to UV light. Keep vials in their boxes or a dark container.
Shelf Life
Once mixed with BAC water, discard the vial after 28 days to ensure sterility and efficacy.
